Abstract: 寒冷な環境に住む変温動物には, 体液を過冷却状態に保ちながら凍結することなく生き延びることが出来るものがいる. これは, 寒冷な環境になると生体内に生成される特殊な不凍糖タンパク質が, 体液中に生成した氷結晶界面に吸着することで, 氷の結晶成長を抑制するためである. 蛍光分子でラベルした不凍糖タンパク質分子の水溶液を冷却し, その中で氷結晶が成長させることで, タンパク質分子と氷結晶との相互作用をその場観察することが初めて可能となった. 本解説では, 氷結晶成長界面でのタンパク質の吸着・拡散挙動, 結晶成長の抑制との関連, 不凍糖タンパク質の界面への2段階可逆吸着など, 不凍糖タンパク質の凍結抑制機構に迫る最新の成果について紹介する.
In situ observations of ice crystal growth in the supercooled solution of antifreeze glycoprotein (AFGP) labeled by the fluorescent molecule were carried out based on the one-directional growth and free growth methods. Diffusion, adsorption, rejection and incorporation processes of AFGP molecules at/in front of the growing interface were clearly observed even at low concentrations. Analysing the fluorescent intensity distribution on each image, we found that the AFGP adsorption proceeded in two steps, namely from weak to strong adsorption states. Furthermore, the adsorption was reversible even under the strong adsorption state. We introduce the recent experimental results and the new adsorption model of AFGP molecules on ice interface.
