Abstract

緑藻クラミドモナスChlamydomonas reinhardtiiは,核,葉緑体,ミトコンドリアの3つのゲノムすべてについて遺伝子導入が可能となっており,外来遺伝子の導入や遺伝子タギング法による突然変異株の作製も容易になっている.本稿では,クラミドモナスの核ゲノムの2種類の形質転換法(エレクトロポレーション法・ガラスビーズ法)を紹介する.
The nuclear transformation techniques are powerful tools for isolation of new mutants, and also for complementation of mutant phenotypes by introducing exogenous DNA segments into the nuclear genome. We described two methods for nuclear genome transformation in a green algae, Chlamydomonas reinhardtii. Linealized DNA fragments encoding selectable markers can be introduced into the nuclear genome by using electroporation and glass beads. Especially dominant selectable markers are useful for the selection of transformants for gene tagging.
