Abstract: The majority of isolates of Aeromonas salmonicida, the causative organism of fish of furunculosis, from diseased fish are rough, that is auto-agglutinating. Therefore serological identification by the slide agglutination test may be difficult to perform. The application of a coagglutination test, for the serological identification of auto-agglutinate A. salmonicida, was studied using staphylococci specifically sensitized with an antibody against the bacterium. This proved to be a simple and rapid method for use in the laboratory and required no special apparatus. The procedure for this method is summarized as follows. 1. A. salmonicida, cultured in nutrient broth, is heated in boiling water for 30min. 2. The supernatant is collected after centrifugation at 4000rpm for 20min. This may be omitted if auto-agglutination is recognized at the bottom of the culture tube. 3. One drop of the supernatant and one drop of anti-A. salmonicida antibody sensitized staphylococci suspension are mixed on a glass slide and the slide is examined after 30min.
特異抗体感作staphylococciを用いたcoagglutination testによるA. salmonicida自然凝集株の血清学的鑑別法について検討を行い次の如き結論を得た。供試菌のブイヨン培養液の加熱抽出液を抗原とするcoagglutination testにより自然凝集株の血清学的鑑別が可能であった。この方法は菌体を含まない加熱抽出抗原を用いるため自然凝集との混同が避けられ,しかも無菌操作を必要としないことから極めて簡便かつ有効な方法である。
