Abstract: Preservation of fish pathogenic viruses under the freezing or freeze-dry conditions was Studied. The viability of viruses after freezing or freeze-dry was estimated. On the freezing method, effects of freezing speed, storage temperatures and cryoprotectants on infectivity of viruses were compared. Fish pathogenic viruses, infectious pancreatic necrosis virus (IPNV), infectious hematopoietic necrosis virus (IHNV), Oncorhynchus masou virus (OMV), Herpesvirus salmonis (H. salmonis) suspended in culture medium supplemented with 10 % DMSO or glycerol were preserved retaining their viabilility under –80 C or liquid nitrogen. DMSO was more effective than glycerol to keep infectivity of virus. Although effect of freezing speed on virus infectivity was not so significant, rapid freezing was more effective than slow freezing. On the freeze-dry method, 2% cryoprotectants, namely lactose and sorbitol, were suitable for IPNV and IHNV. Skimmilk, albumin and sodium glutamate were effective for HMV to avoid the loss of virus infectivity. Storage at ‐20 C or 0 C for the freeze-dried specimens was more effective than at room temperature. Fish pathogenic viruses; IPNV, IHNV, OMV and H. salmonis were preserved at -80 C or in liquid nitrogen and were also preserved by the freeze-dry method.
魚類は変温動物であり,魚類病原ウイルスの至適発育温度は宿主魚類の生息環境水の温度に依存する。たとえばサケ科魚類の病原ヘルペスウイルス Herpesvirus salmonis のように至適発育温度が10Cのものや (26),アメリカナマズのヘルペスウイルスchannel catfish virus (CCV) のように30Cのものも存在する(17)。温度安定性もサケ科魚類の伝染性膵臓壊死症 (infectious pancreatic necrosis virus : IPNV) ウイルスのように低温度下 (0C) で数年間も感染性が保持されるものもあれば (12),サケ科魚類のヘルペスウイルス Oncorhynchus masou virus (OMV) のように0Cでも2週聞程度で感染性を消失するものもある (6)。魚類病原ウイルスの研究を進めるにあたり,ウイルスの性状や病原性を変化させることなく長期間保存することが必要である。しかし,魚類病原ウイルスの保存法についての研究は少ない。本報では魚類病原ウイルスの保存法の開発を目的に,まず低温下における魚類病原ウイルスの温度安定性を検討した。さらに低温保存法における凍害防御剤の種類と濃度,凍結速度および保存温度などのウイルスの安定性に及ぼす影響について検討した。また凍結乾燥保存法における凍害防御剤の種類および保存温度の検討も行ったので,これらの結果を報告する。
