Abstract: 分光測定は,今や生化学,分子生物学では欠かすことのできない基本測定法である.さらに,光合成の分野では光誘起の微少スペクトル変化の測定も,分光光度計を使ってなされる.本項ではこの分光測定の基礎と,その装置の仕組みを解説する.
Principles of spectrophotometry are described. Spectrophotometer is basically composed of three parts; light-source, a monochrometer and a detector. Properties of the three parts are described in detail. Techniques to measure spectra of turbid samples and the samples at 77K are also described. Principles and techniques to determine action spectra and quantum yields are described.
