Abstract: ここではアグロバクテリウムを用いたタバコの核ゲノムの形質転換について紹介する.タバコは葉緑体の形質転換も可能な数少ない植物であり,また大きく育つので生化学実験にも適している.無傷葉緑体も簡単に調製出来る.
Tobacco is a model plant that can be used for both nuclear and chloroplast transformation. In this chapter, we introduce well established methods for Agrobacterium-mediated transformation of the tobacco nucleus. The protocol includes introduction of a binary vector into Agrobacterium, inoculation of tobacco leaf disks with Agrobacterium, and selection and regeneration of transformed tobacco.
