Abstract: Microchip-based immunoassay, which is immunoassay performed on a microchip, has recently been used in various fields owing to its advantages, such as reduction in sample and reagent consumption, short analysis time and simple operation. Different types of immunoassay have been applied to the miniaturization on a microchip. Material and surface modifications of a microchip, introducing and moving samples and detection must be taken into consideration in order to realize a microchip-based immunoassay. Also, the microchip system should be designed according to a type of immunoassay. In the present review article, we explain the types and classifications of immunoassays and describe important points for developing a microchip-based immunoassay. We then introduce several examples of microchip-based immunoassay.
マイクロチップ上で免疫化学反応を行うマイクロチップイムノアッセイは,試料•試薬の微量化,短時間測定,操作の簡便性等の利点により,様々な分野での利用が試みられており,既に多くの種類のイムノアッセイが開発されている.マイクロチップイムノアッセイを実施するには,マイクロチップの材質や表面修飾,サンプルの導入•搬送,検出系といった項目を検討しなければならない.また,マイクロチップ上でイムノアッセイの種類に応じた系を構築する必要もある.本稿では,イムノアッセイの種類と分類について説明するとともに,マイクロチップイムノアッセイを開発する際の要点を論述する.更に,最近の論文で報告されたマイクロチップイムノアッセイの実施例についても紹介する.
