Abstract: 高等生物体内ではL-アミノ酸が過剰に存在して主に利用されており,鏡像異性体の関係にあるD-ア ミノ酸は殆ど存在しない.このL型過剰はホモキラリティと呼ばれ,その偏りの起源解明として地球 外試料中のキラルアミノ酸分析が注目されている.多様な構造異性体を有する様々な有機化合物が含 まれる地球外試料中で微量キラルアミノ酸を分析するためには,高い感度と選択性を兼ね備えた分析 法が必須である.アミノ酸の蛍光誘導体化と複数の分離機構を組み合わせた多次元HPLCは,最も有 用な分析法の一つである.本項では多次元HPLCの開発と様々な試料への適用例,ならびに多次元 HPLCを用いた炭素質隕石中のキラルアミノ酸分析について報告する.
The excess of L-amino acids in higher organisms is called “homochirality,” and its origin is still unclear. Various amino acids have been found in carbonaceous chondrites, and their chiral properties are gathering attention for their use in elucidating chemical evolution, including the origin of homochirality. Highly sensitive and selective multidimensional HPLC systems combining multiple columns of various separation modes are some of the most suitable tools for determining trace amounts of chiral amino acids in extraterrestrial samples, which include various organic compounds with uncountable structural isomers. In this article,the development of a multi-dimensional HPLC system and its application to various real-world samples are described,including an application in chiral amino acid analysis in ca rbonaceous chondrites.
