Hierarchy in the intracellular degradation of proteins. An ER-retarded mutant of bovine AE1 (R664X AE1, red) and the most common mutant of CFTR, ΔF508-CFTR, are degraded by the proteasomal ER-associated degradation (ERAD). Proteasome substrates including various cytosolic proteins such as EGFP (green, upper figures) and ΔF508-CFTR (green, lower figures) form a large cytosolic aggregate, aggresome, at a pericentriolar locus that is indicated by localization of γ-tubulin (blue, upper and lower right figures), whereas R664X AE1 shows no aggresome formation upon proteasome inhibition in transfected HEK293 cells (upper right). However, when cells co-transfected with ΔF508-CFTR (lower left) were exposed to proteasome inhibitors, R664X AE1 is forcibly extracted from the ER to reside in aggresome through association with ΔF508-CFTR (lower right).
